Navepegritide is a prodrug consisting of a CNP moiety (identical to the 38 amino acid sequence of residues 89-126 of endogenous human CNP) transiently conjugated to two branched 20 kDa methoxy polyethylene glycol (mPEG) moieties via a proprietary TransCon Linker. Once administered, the TransCon linker undergoes auto-cleavage to release active CNP, which binds to NPR-B, stimulating the production of cGMP. This inhibits the mitogen-activated protein kinase (MAPK) signaling pathway, thereby antagonizing overactive FGFR3 signaling and promoting chondrocyte differentiation and proliferation to stimulate skeletal bone growth.
