Vusolimogene oderparepvec is a genetically modified HSV-1 oncolytic viral immunotherapy used intratumorally with nivolumab for unresectable advanced cutaneous melanoma refractory to anti-PD-1 therapy, replicating selectively in tumor cells and expressing GALV-GP-R– and GM-CSF to drive anti-tumor immunity. The virus is engineered to express a fusogenic glycoprotein derived from gibbon ape leukemia virus (GALV-GP-R–) and human granulocyte-macrophage colony-stimulating factor (GM-CSF), and has its HSV-1 neurovirulence factor ICP34.5 and antigen-presentation inhibitor ICP47 genes deleted.
