Sugemalimab is an anti-PD-L1 monoclonal antibody used in combination with platinum-based chemotherapy for the treatment of metastatic non-small cell lung cancer. Sugemalimab is a fully human, full-length anti-programmed death-ligand 1 (PD-L1) monoclonal antibody (of the IgG4 isotype) discovered by CStone Pharmaceuticals.

Non-small cell lung cancer (NSCLC) consists of a variety of different lung cancers and is one of the most frequent cancer types. Sugemalimab, a monoclonal antibody indicated as a first-line therapy in combination with chemotherapy for the treatment of metastatic NSCLC, has a dual mechanism of action. The first mechanism involves sugemalimab binding to and targeting the programmed cell death-ligand 1 (PD-L1). The specific binding of sugemalimab to PD-L1 blocks PD-L1 from ligating to programmed cell death protein 1 (PD-1), thereby preventing PD-1/PD-L1 interaction.
The second mechanism involves the antibody inducing antibody-dependent cellular phagocytosis (ADCP) through sugemalimab cross-linking PD-L1-expressing tumor cells with with tumor-associated macrophages without harming effector T-cells.
