Daraxonrasib (RMC-6236) is a noncovalent RAS(ON) multi-selective inhibitor. It binds cyclophilin A and recruits active, GTP-bound RAS to form a ternary complex with KRAS, NRAS, and HRAS, including clinically relevant mutant RAS proteins. This complex occludes effector engagement, suppresses downstream RAF-MEK-ERK signaling, and reduces proliferation of RAS-driven tumor cells.
